Genome evolution: Are microsatellites really simple sequences?
نویسنده
چکیده
In 1989, the world started to look much brighter for genome mappers and behavioral ecologists: the polymerase chain reaction (PCR) and a type of sequence known as a microsatellite were brought together in the development of potentially powerful new methods for mapping genomes. Microsatellites seemed to have almost all the features required by those seeking a simple molecular way of tracking regions of the genome — they are abundant, codominant, distributed over the euchromatic part of the genome and highly polymorphic. At that time, there was no need to know more about the nature of the mutational processes underlying microsatellite variability; it was sufficient to know that microsatellites are stable over a couple of generations. In the subsequent years, microsatellites have significantly influenced both genome mapping and behavioral ecology. The impact of microsatellite analysis on contempory biology is vividly illustrated by the steadily growing number of genes that have been mapped, cloned and characterized by genetic maps based on microsatellites.

MOTIVATION Microsatellites, also known as simple sequence repeats, are the tandem repeats of nucleotide motifs of the size 1-6 bp found in every genome known so far. Their importance in genomes is well known. Microsatellites are associated with various disease genes, have been used as molecular markers in linkage analysis and DNA fingerprinting studies, and also seem to play an important role i...
متن کاملConservation of Human Microsatellites across 450 Million Years of Evolution
The sequencing and comparison of vertebrate genomes have enabled the identification of widely conserved genomic elements. Chief among these are genes and cis-regulatory regions, which are often under selective constraints that promote their retention in related organisms. The conservation of elements that either lack function or whose functions are yet to be ascribed has been relatively little ...
متن کاملUgMicroSatdb: database for mining microsatellites from unigenes
Microsatellites, also known as simple sequence repeats (SSRs) or short tandem repeats (STRs), have extensively been exploited as molecular markers for diverse applications. Recently, their role in gene regulation and genome evolution has also been discussed widely. We have developed UgMicroSatdb (Unigene MicroSatellite database), a web-based relational database of microsatellites present in uni...
متن کاملEvidence for Widespread Convergent Evolution around Human Microsatellites
Microsatellites are a major component of the human genome, and their evolution has been much studied. However, the evolution of microsatellite flanking sequences has received less attention, with reports of both high and low mutation rates and of a tendency for microsatellites to cluster. From the human genome we generated a database of many thousands of (AC)(n) flanking sequences within which ...
متن کاملPatterns of Evolutionary Conservation of Microsatellites (SSRs) Suggest a Faster Rate of Genome Evolution in Hymenoptera Than in Diptera
Microsatellites, or simple sequence repeats (SSRs), are common and widespread DNA elements in genomes of many organisms. However, their dynamics in genome evolution is unclear, whereby they are thought to evolve neutrally. More available genome sequences along with dated phylogenies allowed for studying the evolution of these repetitive DNA elements along evolutionary time scales. This could be...
